Lab outcomes – week July 21

Mon

  • Bought webcam for general lab use; stored in top shelf of left cabinet
  • Raising larave solely on 18-hr brine shrimp test
  1. Started with 300 embryos – 249 larvae hatched, 248 survived
  2. Larvae switched to beakers last Sat (day 4); 18-hr hatched BS feeding started
  3. Larvae not observed eating BS (D4-D6)
  4. Received San Francisco Bay Brine Shrimp – made new cone @ 3.30pm
  5. Continued hatching regular BS @ 18-hr
  • Water change on all medium tanks
  • Switched June 2008 juveniles to glass bowl
  • Discarded 200 larvae saved for algae method – spirulina powder not ready

Tues

  • Water quality test – pH & ammonia
    All big tanks: pH between 5.06-7.37; ammonia level (w/o calculation, obtained from amm. disc) 0.1-0.3
  • Siphoned debris, did 20% water change and cleaned filters on Male T (pH 6.27) & Female T (pH 5.06) tank
  • Excellent spawning: 16/18 tanks, ~3500 embryos
  • ~250 for microinjection, saving the rest
  • Larvae labeled as 072208
  • Larvae raising
  1. SF BS hatched well – fed to larvae
  2. Feeding behavior observed in some larvae; also saw distinct orange bellies (yay)
  3. Larvae count – 247

Wed

  • Pipettes calibrated
  • ~3250 embryos doing well; cleaned and changed water twice
  • Larvae raising – current count 245
  • Lab cleanup – changed table top covers, discarded unsued/old items, reorganized cabinet etc.

Thurs

  • Spawning: 7/11 tanks, ~1700 embryos; 2 big females not releasing eggs
  • Saving all eggs for Ping and June’s experiment next week
  • Larvae labeled as 072408
  • Larvae raising – switched larvae to 1.5L plastic tank; current count 243
  • Lab cleanup – clearing glass vials
  • Paramecia counting attempt using hemocytometer was not good – paramecia were hard to see
  • Sent enquiry to Zfin
  • Also tried with dissection scope – better view – ~50 paramecia/mL

Fri

  • Ongoing experiment: PH#9 Part 1 started – running until Tues
  • Lab cleanup – disposing/cleaning glass vials
  • Larvae 072208 disposed
  • Larvae 072408 used for PH#9 Part 2 & June’s experiment
